A novel ascomycetous yeast Zygoascus doitungensis (family Trichomonasaceae) isolated from soil and rotten wood from Doi Tung Forest, Northern Thailand

Northern Thailand is among the global biodiversity hotspots, and the Doi Tung Forest is recognized as a priority conservation area. During exploration of yeast diversity, three strains representing a novel ascomycetous species were isolated from rotten wood and soil collected in Doi Tung Forest, Chiang Rai Province, Northern Thailand. The combined internal transcribed spacer (ITS) region and the D1/D2 domains of the large subunit (LSU) gene sequence dataset, analysed using maximum likelihood (ML) and Bayesian inference (BI), revealed that isolates represent a new species within the Zygoascus clade, with maximum statistical support (100% ML and 1.00 PP). Zygoascus detingensis was the most closely related species but has over 1.07% sequence divergence in the D1/D2 domains of the LSU gene regions and 1.28% sequence difference in the ITS region. Biochemical and physiological characteristics showed that the utilization patterns of some carbon and nitrogen compounds, and the growth rate at higher temperatures were different compared to Z. detingensis. Therefore, polyphasic characterization, including morphology, phylogeny, and biochemical and physiological characteristics, revealed the novel species Zygoascus doitungensis.
