The present research aimed to develop and evaluate a nanoparticulate composition system (NCS) for the enhanced topical delivery of Luliconazole (LKZ), a broad-spectrum antifungal agent. LKZ-loaded NCS was formulated using a mechanical agitation technique and optimized via Box–Behnken design (BBD), considering drug concentration, lipid concentration, and sonication time as independent variables. The optimized NCS was characterized by particle size, zeta potential, entrapment efficiency (EE), functional, thermal, morphological, and X-ray Diffraction. Transmission Electron Microscopy (TEM), and Scanning Electron Microscopy (SEM) analyses confirmed the formation of uniformly distributed, spherical nanoparticles with smooth surfaces. The optimized LKZ-NCS demonstrated a vesicle size of 360 nm (PDI 0.376), zeta potential of −22.9 mV, and EE of 84.80%. The LKZ-loaded NCS was further incorporated into a Carbopol-based gel and evaluated for pH, rheological properties, and drug diffusion. The results of in vitro permeation study demonstrated regulated drug diffusion from the LKZ NCS gel, compared to conventional and plain formulations, with release kinetics best fitting to zero-order model (R2 = 0.9846). Antifungal activity against Candida albicans, showed a significantly higher zone of inhibition (3.30 ± 0.22 cm2) for the LKZ-NCS incorporated gel, indicating superior efficacy. Stability studies conducted over three months under accelerated conditions revealed insignificant changes in physical appearance and pH, confirming formulation stability. Thus, the developed LKZ-loaded NCS gel presents a promising strategy for effective transdermal delivery of antifungal agents, offering improved drug loading, controlled release, and enhanced therapeutic efficacy.
