Cassava pulp, a starch-industry waste containing high levels of starch and cellulose, has potential as a low-cost alternative carbon source for value-added bioprocesses, such as amino acid fermentation. This research aimed to produce glucose from cassava pulp using enzymatic hydrolysis. The key parameters investigated in a 250 mL flask were temperature, pH, average particle size, pretreatment, and concentration of cassava pulp via enzymatic hydrolysis in a 250 mL flask. The agitation speed and hydrolysis time of the enzymes were studied for preliminary reactor validation in a 500 mL reactor. In a 250 mL Flask, the optimal conditions of cellulase enzyme were initially examined, followed by α-amylase and amyloglucosidase. The optimal temperature and pH for cellulase were 60°C and 5.5, for α-amylase were 90°C and 6.0, and for amyloglucosidase were 60°C and 6.0, respectively, serving a maximum glucose concentration of 28.2 ± 1.0 g/L. Subsequently, the residue pulp after enzymatic hydrolysis was pretreated by immersing it in a 2 wt% NaOH solution. The pretreated pulp showed an optimal temperature of 55°C and an optimal pH of 5.0 when using cellulase, resulting in a glucose concentration of 6.22 ± 0.13 g/L. The average particle size influenced the glucose concentration, as confirmed by ANOVA analysis. Moreover, gelatinization also enhanced enzymatic hydrolysis. The optimal concentration of cassava pulp was found to be 5 wt%, which resulted in a higher glucose concentration at 32.5 ± 0.2 g/L. In a 500 mL reactor, an agitation speed of 400 rpm was selected to determine the optimal hydrolysis time. The hydrolysis time for cellulase was 24 h, α-amylase was 2 h 30 min, while for amyloglucosidase it was 3 h 30 min, resulting in a glucose concentration of 37.3 ± 0.9 g/L. For cellulase, when using pretreated residue pulp as the raw material, the glucose concentration reached 8.22 ± 0.40 g/L after 90 h of hydrolysis. Therefore, a sequential enzymatic hydrolysis with secondary valorization via residual pulp hydrolysis glucose has a glucose yield of 0.727 ± 0.033 and a higher saccharification efficiency of 93.7 ± 3.4%. This glucose solution serves as an economical carbon substrate for industrial applications, demonstrating an effective multi-scale framework for starch waste valorization and cost reduction.
