Poly(3-hydroxybutyrate) production from sugarcane bagasse by Paracoccus sp.: Kinetic modeling using modified logistic and Luedeking–Piret models incorporating cell death and PHB consumption

This study aimed to enhance poly(3-hydroxybutyrate) (PHB) production from sugarcane bagasse (SB) hydrolysate via fed-batch fermentation using Paracoccus sp. KKU01. Following alkaline hydrogen peroxide pretreatment, 121 g/L of pretreated SB was enzymatically saccharified, yielding a hydrolysate containing 103.0 ± 0.8 g/L of reducing sugar. Batch fermentation using this hydrolysate yielded 8.3 ± 0.0 g/L of PHB, with a PHB content, productivity, and yield of 22.7 ± 0.6% (w/w), 0.20 ± 0.01 g/(L·h), and 0.14 ± 0.01 g/g, respectively. Fed-batch fermentation increased the PHB titer by 70% to 14.1 ± 1.4 g/L and PHB content to 31.6 ± 4.2% (w/w), achieving a productivity and yield of 0.21 ± 0.02 g/(L·h) and 0.14 ± 0.01 g/g, respectively. While the classical logistic and Luedeking–Piret models were inadequate for describing changes in biomass and PHB profiles, incorporating growth decay and PHB consumption terms considerably improved their accuracy, enabling the prediction of biomass decline and PHB reduction. The modified models were validated using several metrics, including R 2 , root mean square error, and the corrected Akaike Information Criterion. The achieved PHB titer is among the highest reported for SB hydrolysate, and the proposed modified models successfully capture late-stage fermentation kinetics overlooked in the classical models.
