Indigenous insect-associated bacteria may provide biological resources for improving disease resilience in insect production. We isolated and functionally screened gut-associated bacteria from the Thai polyvoltine Nang Lai strain of Bombyx mori. Fifty colonies were recovered following alkaline enrichment. Partial 16S rRNA gene sequence analysis and phylogenetic comparison assigned isolates KI1, KI2, KI3, KI4, and KI9 to Staphylococcus gallinarum and KI6 to Enterococcus mundtii; the sequences were deposited in GenBank under accession numbers PZ319142-PZ319147. KI2 and KI4 produced inhibition zones against Bacillus cereus, Bacillus thuringiensis, and Serratia marcescens, whereas KI6 and KI9 showed no detectable inhibition. KI4 was selected for in vivo evaluation because it produced the largest inhibition zone against B. cereus (18.0 ± 1.1 mm). Under non-challenged conditions, KI4 supplementation did not differ significantly from the untreated control in final larval weight, cocooning, pupation, fresh cocoon weight, or shell weight. B. cereus challenge reduced endpoint survival to 49.33 ± 2.31%, whereas daily KI4 supplementation initiated before challenge and continued thereafter was associated with higher endpoint survival (77.33 ± 2.31%) and higher cocooning and pupation than the pathogen-only treatment. The in vitro antagonism and in vivo host response represent separate observations because a non-antagonistic bacterial control was not included. These findings support further evaluation of S. gallinarum KI4 (GenBank accession no. PZ319145) as a candidate host-derived bacterial resource for sericulture. Intestinal persistence, the mechanism and specificity of the host response, biosafety, and field efficacy require further investigation before practical application.
