Information from the abstract

Abstract Paragonimiasis is endemic in the Northeastern states of India. The study is aimed to isolate the Paragonimus westermani metacercariae from the freshwater crabs collected at two endemic areas Moreh and Churachandpur in Manipur, and to determine the species by morphological and molecular characterization of the metacercariae. A total of 300 freshwater crabs were collected, 140 from Churachandpur and 160 from Moreh were identified as Maydelliathelphusa lugubris and Alcomon superciliosum respectively. The crab extract was prepared by artificial digestion, fractional filtration and sedimentation. P. westermani like metacercariae were isolated by observing under the stereo dissecting microscope. Altogether 45 P. westermani like metacercariae twenty from Churachandpur and twenty-five from Moreh were isolated. They were grouped separately for morphological and molecular study. Four metacercariae from each group were preserved in 80% alcohol for DNA extraction, while the remaining metacercariae were fixed in 10% formalin for morphological study. Morphological characterization of the metacercariae was conducted under a stereo zoom dissecting microscope. For molecular identification, genomic DNA was extracted from individual metacercariae and subjected to PCR amplification and sequencing targeting the internal transcribed spacer 2 and mitochondrial cytochrome c oxidase subunit 1 genes, followed by phylogenetic analysis. Morphological characterization and molecular analysis of the specimens confirmed their identity as P. westermani . Phylogenetic analysis based on ITS2 and cox1 sequences revealed two distinct clades, with ITS2 showing close similarity to the isolates from other Northeastern states of India.
