Unveiling potential natural promoters of calcium oxalate kidney stones in the urine via anion-exchange chromatography, crystal assays, and proteomics

Siriraj researchers fractionated stone-former urine by anion-exchange chromatography, assayed crystallization, growth, aggregation and cell adhesion, and proteomically profiled highly promoting fractions. Four proteins recurred as candidates, not yet confirmed causal stone promoters.

Key findings

  • Nearly all fractions promoted at least one crystal process; SFQ2, SFQ8 and SFQ9 scored highest and contained 12, 38 and six proteins. CD44, galectin-3-binding protein, kallikrein-1 and AMBP appeared in more than one fraction.

Why this matters globally

Recurrent calcium-oxalate stones need better mechanistic targets. Natural urinary promoters may yield biomarkers or therapies, but each protein's role must be separated from mixture effects.

Seven Siriraj Hospital and Mahidol University researchers conducted the urine, crystal and mass-spectrometry work, making Thailand the study center.

Limitations to consider

Sample size, controls and person-level variation are absent from the abstract. Fractions contain mixtures and the summed score needs validation; recurrence does not prove causation, with no purified-protein, knockdown, animal or validation-cohort evidence.
