Multi-Targeted Anti-Alzheimer’s Effects of Tri-Sannibat-Phol: Biological Evaluation, Behavioral Validation, and LC-MS/MS Phytochemical Profiling

Researchers evaluated the Thai polyherbal formula Tri-Sannibat-Phol in antioxidant, acetylcholinesterase and amyloid-beta assays, followed by neuronal-cell and scopolamine mouse models. Several outcomes improved and piperine was a prominent constituent, but the evidence remains entirely preclinical.

Key findings

  • The extract improved cell viability across 1-100 micrograms/mL, had a reported acute LD50 above 2,000 mg/kg, and medium-to-high doses reversed several behavioural deficits. LC-MS/MS identified 13 piperidine alkaloids, with piperine at 17.61±0.80% w/w.

The study systematically links traditional knowledge with chemical profiling and multi-level models, providing a foundation for active-component isolation, product standardisation and chronic-toxicity work before human trials are considered.

Limitations to consider

The scopolamine model does not reproduce the full pathology of Alzheimer's disease, toxicity testing was acute only, and a multicomponent formula prevents attribution to one compound or mechanism. Human efficacy, pharmacokinetics, interactions and safety are unknown.
