FREEZE-DRIED BINARY AND TERNARY SOLID DISPERSIONS OF CANNABIDIOL: FORMULATION, CHARACTERIZATION, AND SOLUBILITY ENHANCEMENT

A Naresuan University team developed amorphous solid dispersions of cannabidiol using PVP and surfactants. Optimised ternary formulations containing poloxamer 407 increased solubility by about 11,000-fold, dissolved rapidly and retained amorphous character with over 96% CBD after six months. This solves a formulation problem, not human bioavailability, efficacy or safety.

Key findings

  • Binary dispersions increased solubility up to 32-fold; initial ternary screening reached about 7,000-fold, led by poloxamer 407. Optimised F7/F14 reached roughly 11,000-fold with rapid dissolution. DSC supported amorphisation, FTIR indicated intermolecular interactions, and over 96% CBD remained after six months with amorphous character.

Why this matters globally

The platform may support more consistent, rapidly dissolving CBD products and other poorly water-soluble compounds, provided residual solvent and recrystallisation are controlled.

Limitations to consider

Fold changes are reported without absolute solubility or full dissolution conditions. Permeability, pharmacokinetics, food effects, dose, toxicity and efficacy are absent. Residual tert-butyl alcohol and stability across humidity and temperature require assessment.
