CXCR2 blockade potentiates HDAC3 deletion mediated antitumor immunity in colorectal cancer

HDAC3 is highly expressed in colorectal cancer (CRC) and associated with poor prognosis. This study reveals that HDAC3 deletion dually regulates chemokine expression through epigenetic mechanisms, promoting CD8⁺ T cell infiltration (via CXCL10) on one hand, while enhancing the recruitment of myeloid-derived suppressor cells (MDSCs) (via CXCL1/2/3/5) on the other. The latter, mediated through CXCR2 signaling, counteracts the intrinsic anti-tumor effect of HDAC3 deletion. We further demonstrate that combining a CXCR2 inhibitor effectively blocks MDSC infiltration and significantly enhances the immunotherapeutic efficacy mediated by HDAC3 deletion. This study systematically elucidates the "double-edged sword" role of HDAC3 in the CRC immune microenvironment and provides a theoretical and experimental basis for an HDAC3/CXCR2 dual-targeting therapeutic strategy.

The Asia Pacific League of Associations for Rheumatology ( APLAR ) Consensus Statements for the Management of Reproductive Issues in Autoimmune Rheumatic Diseases

BACKGROUND: The management of reproductive health in individuals with autoimmune rheumatic diseases (AIRDs) has evolved into a primary clinical priority. Wide variations in clinical practice and drug availability in the Asia-Pacific regions necessitate localized frameworks. These consensus statements, developed by the Asia Pacific League of Associations for Rheumatology (APLAR) Special Interest Group for Women's Health & Reproductive Issues in Rheumatic and Musculoskeletal Diseases, aim to bridge the gaps within the region, foster multidisciplinary collaboration, and address regional knowledge deficits. METHODS: An expert panel comprising 16 members of the 13 APLAR member nation organizations formulated 23 research questions using the PICO framework. A systematic review of English-language literature up to July 2025 was conducted across the MEDLINE, Scopus, Google Scholar, and Cochrane Library databases, supplemented by a manual search of journals from the APLAR regions. Evidence from the APLAR region was appraised using the GRADE system. A modified Delphi online voting process was employed to reach consensus, pre-defined as ≥ 75% agreement. RESULTS: The panel approved four overarching principles and 86 statements, organized into 16 broad categories. These statements cover all three phases of pregnancy in AIRDs, the safety of medications during pregnancy and lactation, contraception, assisted reproductive techniques, fertility preservation, and hormonal replacement therapy, covering both female and male aspects as relevant. CONCLUSION: In a field often lacking high-quality data, these consensus statements from APLAR provide expert opinion-based guidance to support clinical decision-making. GRB 241030A: A bright afterglow challenging forward shock emission

Context. Gamma-ray burst GRB 241030A ( z = 1.411) exhibited a particularly bright afterglow (similar to the ‘BOAT’, GRB 221009A), detected across gamma-ray, X-ray, UV, and optical bands. The extensive, multi-wavelength observations of this remarkable event provide a valuable opportunity to advance our understanding of GRB afterglow physics. Aims. We aim to constrain the physical properties of the jet, its microphysics, and the characteristics of the circumburst environment in the context of forward-shock emission. Methods. We compiled multi-wavelength observations spanning from a minute to a week after the prompt emission, processing the data through a unified photometry pipeline. Leveraging this comprehensive dataset, we analysed the observations both analytically and using Bayesian inference with two independent models. Our models assume that the afterglow emission arises from the strong forward shock of a laterally structured jet, with possible contributions from synchrotron self-Compton (SSC) scatterings. Results. We find that our models do reproduce the afterglow observations accurately, from the X-rays to the optical, favouring a jet propagating into a constant-density interstellar medium, with a viewing angle within the jet core. However, both analyses – with and without the inclusion of SSC scatterings – require parameter values that are extreme compared to expectations from standard theory. In particular, our results imply extremely energetic jets despite regular prompt energy, leading to a very inefficient prompt emission. Furthermore, the jets are particularly inefficient at accelerating particles, with low ϵ e and ϵ B , leading to significant SSC emission. Pioneering plant virology research for a healthier future: a summary of the 2025 American Society for Virology (ASV) Plant Virology Satellite Symposium

The American Society for Virology (ASV) 2025 Annual Meeting, held from 14 to 17 July at the Palais des congrès de Montréal and hosted by McGill University, featured a dedicated Plant Virology Satellite Symposium entitled "Pioneering Plant Virology Research for a Healthier Future." This symposium emphasized the inclusion of topics of significance to early-career scientists. The program highlighted advances across diverse areas of plant virology, including mechanisms of transgenerational virus transmission, the economic and biological impacts of grapevine viruses on viticulture, emerging plant virus vectors, and innovative applications of plant viruses as nanomaterials for cancer immunotherapy. Additionally, emerging research on small functional proteins encoded by plant viruses and plant defense mechanisms underscored the complexity of viral genomes and host interactions. The symposium showcased cutting-edge approaches and strategies aimed at addressing global challenges in plant health and food security.

Global disparities in hearing care services and infrastructure: findings from a 47-country international provider survey

OBJECTIVE: Access to audiological services and support for individuals with hearing loss varies widely across low- and middle- income countries (LMICs) and high-income countries (HICs). This study examines disparities in the availability of audiological services across World Bank income groups. DESIGN: An international cross-sectional survey was developed and distributed by the Global Otolaryngology Head and Neck Surgery Initiative. The survey evaluated the availability of audiology and support services across countries. Multiple responses from one country were combined into one entry. Statistical significance between groups was assessed using chi-squared tests.Study Sample: A total of 135 responses were received from 47 countries (30 LMICs and 17 HICs). RESULTS: Significant disparities were identified across most service categories. HICs reported greater availability of newborn hearing screening, diagnostic tests, paediatric and adult hearing assessments, vestibular services, and hearing technologies compared to LMICs. Advanced diagnostic tools and hearing devices such as cochlear implants were significantly more accessible in HICs. CONCLUSIONS: Substantial differences were observed in audiological and rehabilitative services across HICs and LMICs. Efforts to bridge significant gaps are essential for achieving equitable hearing health care worldwide.

Stapokibart provides significant improvements in signs and symptoms of atopic dermatitis irrespective of prior systemic treatment: a post-hoc analysis of a phase 3 trial

Objectives This post-hoc analysis of CM310AD005 conductedaimed to analyze the efficacy and safety of stapokibart in adults with moderate-to-severe atopic dermatitis (AD) with and without prior systemic treatment.Methods In CM310AD005, eligible patients were randomized 1:1 to receive stapokibart 600 (loading dose)-300 mg and placebo Q2W for 16 weeks; all patients subsequently received stapokibart 300 mg Q2W for 36 weeks.Results At week 16, in systemic treatment-naïve patients, stapokibart led to significantly higher response rates than placebo for ≥75% improvement from baseline in the Eczema Area and Severity Index score (EASI-75, 70.3% vs. 29.1%), EASI-90 (38.1% vs. 13.3%), Investigator’s Global Assessment score of 0 or 1 (45.2% vs. 19.4%), and ≥4-point reduction in weekly average of daily Peak Pruritus Numerical Rating Scale score (34.8% vs. 13.9%) (all p < 0.0001). Among patients with prior systemic treatment, these response rates were also significantly higher in the stapokibart group, reaching 61.5% vs. 19.3%, 35.4% vs. 7.2%, 42.7% vs. 9.6%, and 37.5% vs. 7.2% (all p < 0.0001), respectively. All patients showed further improvements through week 52. The most common treatment-emergent adverse events were infections and infestations, occurring in 61.6% and 70.5% of patients with and without prior systemic treatment.Conclusions Stapokibart demonstrated significant clinical efficacy and manageable safety in AD patients irrespective of prior systemic treatment.

Resolving CYP2D6 Structural Complexity with Long‐Read Sequencing: Implications for Tamoxifen Precision Dosing in Thai Breast Cancer Patients

Cytochrome P450 2D6 (CYP2D6)-mediated bioactivation of tamoxifen to endoxifen is a key determinant of therapeutic efficacy in estrogen receptor-positive breast cancer. In Asian populations, the high prevalence of structurally complex CYP2D6 alleles complicates conventional genotyping, limiting accurate genotype-phenotype correlation. To address these gaps, long-read Oxford Nanopore sequencing was applied to resolve CYP2D6 structural complexity and evaluate its impact on endoxifen metabolism. The study cohort included 492 Thai breast cancer patients, with multivariable analysis performed on 361 individuals receiving standard-dose tamoxifen to determine the association between CYP2D6 activity scores and plasma endoxifen concentrations. Long-read sequencing identified 82 distinct diplotypes and successfully resolved complex structural variations, including the prevalent CYP2D6*36 + *10 non-identical duplication. Overall, 46.55% of patients were classified as having decreased enzyme function, primarily intermediate metabolizers (45.53%). CYP2D6 activity score emerged as the strongest determinant of endoxifen exposure (P < 0.001). Decreased-function genotypes were significantly associated with lower endoxifen levels (using an exploratory benchmark of ≤5.97 ng/mL), independent of parent drug concentration. These findings demonstrate that long-read sequencing enables the precise resolution of CYP2D6 structural complexity. By accurately capturing rare and complex CYP2D6 alleles, the results indicate that standard genotyping may misclassify certain patients, potentially placing a large portion of the studied population at an unrecognized risk of low tamoxifen exposure.

Building sustainable workplaces: The role of organizational climate and recognition on employee task performance in Vietnamese SMEsThis research aims to study the relationships between Organizational Climate and Task Performance through Organizational Recognition in the context of small and medium-sized enterprises (SMEs) in Da Nang, Vietnam. Structural Equation Modeling (SEM) is used to examine the direct and indirect (mediated) relationships among the independent variables, the mediator, and the dependent variable. The population of this study is individual employees in SMEs; 402 respondents were obtained through a convenience and volunteer sampling method. The empirical results demonstrated an excellent model fit. The path analysis revealed that all four dimensions of Organizational Climate had significant positive direct effects on Organizational Recognition (p < 0.001), and that Organizational Recognition was significantly associated with Task Performance. Bootstrap analysis with 5000 resamples and bias-corrected 95 % confidence intervals indicated that all four indirect effects through Organizational Recognition were statistically significant. Standard of Work was fully mediated by Organizational Recognition, whereas Organizational Structure, Responsibility, and Commitment were partially mediated, retaining significant direct associations with Task Performance. These results suggest that, in the SME context of Da Nang, Organizational Recognition is an important mechanism through which a supportive climate is associated with employee performance. The study provides insights for SME management seeking to strengthen recognition practices as part of a more humane and socially sustainable work environment.
